Abstract:
In this study, the optimum conditions for hydrostatic induction of triploid
Leuciscus brandti were selected by means of histological and comparative tests. The results showed that: (1) The tissue section examined 5min after fertilization showed that the second polar body was obviously protruding from the egg membrane of the fertilized egg, indicating its readiness for discharge. (2) The fertilization rate of 5min initiation time was not significantly different from the control group (
P>0.05), but was significantly higher than that of other experimental groups (
P<0.05). There was no significant difference in incubation rate between 4min and 5min (
P>0.05), but it was significantly lower than that of the control group (
P<0.05), and significantly higher than that of other experimental groups (
P<0.05). The triploid rate of the initial time of 5min was significantly higher than that of other experimental groups (
P<0.05). (3) There was no significant difference between the fertilization rate of hydrostatic pressure 60 MPa and control group (
P>0.05), but it was significantly higher than that of other experimental groups (
P<0.05). The hatching rate of 65 MPa was significantly lower than that of other groups (
P<0.05), and the hatching rate of 50, 55 and 60 MPa had no significant difference (
P>0.05), but was significantly lower than that of the control group (
P<0.05). The triploid rate of 60 MPa was significantly higher than that of other experimental groups (
P<0.05). (4) There was no significant difference in fertilization rate between 4 and 5min induction duration (
P>0.05), but it was significantly higher than that of other experimental groups (
P<0.05), and significantly lower than that of the control group (
P<0.05). The hatching rate of experimental groups was significantly lower than that of the control group (
P<0.05), and the hatching rate and triploid rate lasting 4min were significantly higher than those of other experimental groups (
P<0.05). Under the conditions of this experiment, with a water temperature of (17±1)℃, the best induction conditions for triploid
Leuciscus brandti were 5min after fertilization, the hydrostatic pressure of 60 MPa, and the continuous treatment duration of 4min.
